Abstract
Se describe un método para la preparación de placas de inmunodifusión radial para la cuantificación de la lgG humana. El método se basó en las siguientes etapas: (1) aislamiento de lgG de suero normal
mediante cromatografía de afinidad; (2) inmunización en cobayo con lgG completa en un esquema breve para evitar la inducción de anticuerpos anti-cadenas livianas; (3) evaluación de la especificidad del antisuero; (4) preparación de las placas y estandarización de las variables; y (5) evaluación del
método en estudio frente a un método comercial usado corno referencia. La correlación obtenida fue de 0,93 (p<0,01) lo que nos permite concluir que el método es adecuado, y podría implementarse a escala mayor con fines de producción.
A method for the preparation of radial immunodiffusion plates for the quantitation of hum an lgG is described. It is based upon the following steps: (1) isolation of IgG from normal serum by affinity chromatography; (2) immunization of guinea pigs using a short schedule to avoid response against light chains; (3) assessment of antiserum specificity; (4) preparation of pIates and standardization of variables; and, (5) evaluation of the proposed method by a correlation analysis with a commercially - available method utilized as reference. The correlation obtained was 0.93 (p<0.O1), indicating that the method is adequate, providing a basis for a future large - scale production.
